| 内容記述 | Escherichia coli and Salmonella enterica are Gram-negative bacteria. Some strains of both bacteria can cause severe diseases for animals including humans. Ralstonia solanacearum is a soil-borne gram-negative phytopathogenic bacterium causing bacterial wilt disease in many important crops such as rice and tomato. The abuse of antibiotics for treating those pathogens leads to economical effect and increase antibiotic resistance. Bacteriophages (or phages) are viruses which infect and kill specific kind of bacteria with high host specificity. Phage therapy, to cure bacteria infectious diseases using phages, is gaining attention as one of the alternative treatments for bacterial infections. In chapter 1, I isolated 4 kinds of bacteriophages, one is Ralstonia solanacearum phage φRS138 and the three are E. coli phages KIT01, KIT02, and KIT03. The morphological observation of these phages by TEM indicated that KIT01 and KIT03 belong to the family Myoviridae while KIT02 and φRS138 belong to the family Siphoviridae. Genome analysis showed φRS138 is the characteristic of temperate phage with high identity to plasmids of some R. solanacearum. In chapter 2, I analyzed the host range of Escherichia coli phages KIT01, KIT02, and KIT03. These are very unique since they lyse both Salmonella and E. coli. I also analyzed the genome sequence of KIT03 and performed genome annotation and alignment. In chapter 3, I analyzed the phage growth parameters of KIT01, KIT02, and KIT03. The in vitro application of these phages were evaluated to suppress the growth of E. coli and Salmonella. KIT01, KIT02 and KIT03 can cause the bacteriolysis in single apply and synergic effect in cocktail against some strains of E. coli and Salmonella. I further revealed that KIT01 and KIT03 recognizes and adsorbs lipopolysaccharide and outer membrane protein A of bacteria. KIT02 recognizes and utilize BtuB as s receptor with the support of lipopolysaccharide. In conclusion, KIT01, KIT02, KIT03, and these mixtures can be used for controlling both E. coli and Salmonella. | |||||||||
